Ván cờ Trịnh Nguyễn được chơi bởi hai người, người cầm quân Nguyễn (Xanh) đi trước, người cầm quân Trịnh (Đỏ) đi sau. Mục tiêu của ván cờ là [[Chiếu hết (cờ vua)|chiếu hết]] quân [[Chúa Trịnh|Trịnh]] hoặc [[Chúa Nguyễn|Nguyễn]] của đối phương.

Có tất cả 32 quân cờ chia đều cho hai bên (xanh/đỏ), mỗi bên có 16 quân (6 loại quân) bao gồm 1 Chúa (Trịnh/Nguyễn), 2 Vệ, 2 Tượng, 2 Mã, 2 Xe và 7 Tốt.
Chúa được xếp ở cột thứ 5 - hàng đầu tiên, bên trái và bên phải Chúa là 2 Vệ, ra ngoài là 2 Tượng rồi đến 2 Mã và 2 Xe nằm ở 2 cột ngoài cùng. Các Tốt được xếp ở các cột 1,2,4,5,6,8,9 - hàng thứ 3 mỗi bên.
